The vertebrate homologs of Hh comprise of Sonic (SHh), Desert (DHh) and Indian (IHh) hedgehogs. They play important roles in embryonic development and in diseases such as cancer. Human SHh precursor is a 45 kDa protein. An autocatalytic reaction yields a 19 kDa N-terminal domain that is biologically active. Because of unique post-translational modifications, SHh produced in mammalian cells is much more active and, importantly, more biologically authentic than SHh produced from E. coli. StemRD expresses and purifies human SHh from a human cell line in animal-free medium. This method avoids potential artificial effect caused by bacterial product known to present in commercial SHh made from E. coli (1). Through a novel and proprietary process, the protein is purified to near homogeneity. Data shown below is from a typical experiment when compared to an E. coli product on the market.
